Fludarabine Phosphate, Cyclophosphamide, Total Body Irradiation, and Donor Stem Cell Transplant...
Accelerated Phase Chronic Myelogenous LeukemiaBCR-ABL1 Positive38 moreThis phase II trial studies how well fludarabine phosphate, cyclophosphamide, total body irradiation, and donor stem cell transplant work in treating patients with blood cancer. Drugs used in chemotherapy, such as fludarabine phosphate and cyclophosphamide, work in different ways to stop the growth of cancer cells, either by killing the cells, by stopping them from dividing, or by stopping them from spreading. Radiation therapy uses high energy x-rays to kill cancer cells and shrink tumors. Giving chemotherapy and total-body irradiation before a donor peripheral blood stem cell transplant helps stop the growth of cells in the bone marrow, including normal blood-forming cells (stem cells) and cancer cells. It may also stop the patient's immune system from rejecting the donor's stem cells. When the healthy stem cells from a donor are infused into the patient they may help the patient's bone marrow make stem cells, red blood cells, white blood cells, and platelets. The donated stem cells may also replace the patient?s immune cells and help destroy any remaining cancer cells.
A Long-term Safety and Efficacy Study of Danicopan as an Add-on Therapy to Complement Component...
Paroxysmal Nocturnal HemoglobinuriaThis is a single-arm long-term extension study that will enroll participants with PNH who have completed participation in Alexion-sponsored clinical studies with danicopan as an add on therapy to a C5i.
A Long Term Safety Study of BCX9930 in Subjects With Paroxysmal Nocturnal Hemoglobinuria (PNH)
Paroxysmal Nocturnal HemoglobinuriaPNHThis study is designed to evaluate the long-term safety of daily oral treatment with BCX9930 in subjects who have participated in a previous BCX9930 trial for PNH and showed a benefit of treatment as determined by the Investigator. The study allows continued access to BCX9930 for enrolled subjects. The study will also evaluate the long-term effectiveness and impact on quality of life and general well-being of BCX9930 treatment, and the subject's satisfaction with the medication.
A Phase III Study Evaluating The Efficacy And Safety Of Crovalimab Versus Eculizumab In Participants...
Paroxysmal Nocturnal HemoglobinuriaA study designed to evaluate the non-inferiority of crovalimab compared with eculizumab in participants with PNH who have not been previously treated with complement inhibitor therapy. This study will enroll approximately 200 participants.
Study of the Oral Factor D (FD) Inhibitor ALXN2050 in PNH Patients as Monotherapy
Paroxysmal Nocturnal Hemoglobinuria (PNH)The study will evaluate the efficacy and safety of the oral Factor D (FD) inhibitor ALXN2050 (ACH-0145228) monotherapy in patients with PNH that are treatment naïve, or patients currently treated with eculizumab who still experience anemia and reticulocytosis, or patients currently treated with ALXN2040 (danicopan) as monotherapy. After signing consent, participants will have periodic visits through Week 12, at which time the primary endpoint and key secondary assessments will be analyzed. Participants will continue on treatment past 12 weeks into a long-term extension portion of the trial.
Pozelimab and Cemdisiran Combination Treatment in Adult Participants With Paroxysmal Nocturnal Hemoglobinuria...
Paroxysmal Nocturnal HemoglobinuriaThe primary objective of the study is to evaluate the safety and tolerability of 2 dosing regimens of pozelimab and cemdisiran combination therapy during the open-label treatment period (OLTP) The secondary objectives of the study are: To evaluate the effect of the combination treatment on the following parameters of intravascular hemolysis: lactate dehydrogenase (LDH) control, breakthrough hemolysis, and inhibition of total complement hemolysis activity (CH50) To evaluate the effect of the combination treatment on hemoglobin levels To evaluate the effect of the combination treatment on red blood cell (RBC) transfusion requirements To evaluate the effect of the combination treatment on clinical outcome assessments (COAs) measuring fatigue and health related quality of life To assess the concentrations of total pozelimab in serum and total complement component (C) 5 and cemdisiran in plasma To assess immunogenicity to pozelimab and cemdisiran To evaluate the long-term safety and efficacy of pozelimab and cemdisiran in an optional open-label extension period (OLEP) To assess safety after treatment intensification with pozelimab and cemdisiran
BCX9930 for Treatment of PNH in Subjects With Inadequate Response to C5 Inhibitor Therapy
Paroxysmal Nocturnal Hemoglobinuria (PNH)The purpose of this study is to determine the efficacy and safety of BCX9930 monotherapy for the treatment of PNH compared to continued C5 inhibitor therapy in adult PNH patients with residual anemia despite treatment with a C5 inhibitor.
BCX9930 for the Treatment of PNH in Subjects Not Receiving Other Complement Inhibitor Therapy
Paroxysmal Nocturnal Hemoglobinuria (PNH)The purpose of this study is to determine the efficacy and safety of BCX9930 monotherapy for the treatment of adult patients with PNH not currently receiving complement inhibitor therapy.
