The purposes of this study are to: Learn about the safety of CK-3773274 after a single dose and multiple doses in healthy subjects. Learn how healthy subjects tolerate CK-3773274 after a single dose and multiple doses. Find out how much CK-3773274 is in the blood after a single dose and multiple doses. Determine the effect of doses of CK-3773274 on the pumping function of the heart. Evaluate the effect of cytochrome genetic variants on how the body metabolizes CK-3773274. Evaluate the effect of a meal on how much CK-3773274 is in the blood in healthy adult subjects. Evaluate whether the amount of CK-3773274 in the blood is the similar for both the tablet and granules in capsule forms of the drug.

The purpose of RHYME is to evaluate the safety and efficacy of ranolazine in Hypertrophic Cardiomypathy patients with chest pain or dyspnea despite treatment with standard medical therapy. This is a small, pilot, open-label (non-randomized) study of an approved drug for the treatment of angina in a novel patient population (adult patient population with hypertrophic cardiomyopathy).

Study of Exercise Training in Hypertrophic Cardiomyopathy

The investigators propose a pilot randomized controlled trial to determine the safety and potential benefits of moderate intensity exercise in patients with hypertrophic cardiomyopathy. The investigators hypotheses are that exercise parameters derived from a baseline cardiopulmonary exercise test will target an appropriately safe level of exercise intensity that will not cause significant arrhythmias or exacerbate symptoms and that exercise training for 4 months will result in significant improvements in peak oxygen consumption (peak VO2) and quality of life, with neutral effects on the clinical characteristics.

The purpose is to examine the effects of time-restricted eating (TRE; consuming all calories within an 8-hour period each day) vs. normal eating (CON; consuming same kcals and protein as TRE, but during a 10-13 hr eating window each day) during 8 weeks of resistance exercise on body composition and muscle mass (whole muscle and single fiber), muscular performance, anabolic protein signaling, single muscle fiber characteristics, and the gut microbiome in well-trained young men and women.

Skeletal Muscle Hypertrophy

When you perform weightlifting, your body makes new proteins within your muscle. These new proteins can increase the size of the fibers within your muscle to make your muscle larger, a process called hypertrophy. The common convention surrounding gains in muscle mass and strength are that higher-loads (i.e. heavier weights) used for fewer repetitions are better for increasing strength and lower-loads (i.e. lighter weights) used for higher repetitions are better for increasing muscle mass. However, recent research has found that when higher- and lower-loads are used when participants exercise until volitional fatigue (i.e. cannot perform another repetition), muscle mass and strength increases are similar regardless of using a higher- or lower-load. Many of these studies have examined this effect in males with fewer studies examining the effects of higher- and lower-load training in females when assessing changes in muscle mass, strength, and muscle endurance. Further, it has been shown that there is substantial individual variation in response to resistance exercise training where individuals can be broadly categorized as higher- or lower-responders to resistance exercise training. This study aims to explore how the muscle mass, strength, and muscle endurance of females are impacted by both higher- and lower-loads while also exploring how individuals may respond to the training interventions.

Hypertrophic Cardiomyopathy

To explore the feasibility, safety, health and psychological benefits of a 12-week high intensity exercise programme in a young group of individuals with hypertrophic cardiomyopathy (HCM). This will pave the way for a large-scale randomised study of safety of exercise in HCM, the results of which will strengthen the evidence base for exercise recommendations.

The Effect of Insect Protein on Performance and Hypertrophy

Protein Supplementation

Purpose: to study the effect of insect protein on performance and hypertrophy during 8 weeks of resistance training. Hypothesis: Insect protein supplementation enhances the effect of resistance training on muscle mass and muscle strength. Primary outcome: Muscle hypertrophy and muscle strength Design: Two groups of healthy young men (age: 18-30 years) with resistance training experience, but no structural resistance training performed for the last 12 months. Randomized controlled intervention study +/- insect protein (placebo group ingest isocaloric carbohydrate). Both groups perform 8 weeks of progressive resistance training (4/week) aiming to induce muscle hypertrophy.
