Glucose Metabolism DisordersThis project investigated how glucose metabolism differs due to sleep deprivation for three consecutive nights as compared to sufficient sleep for three nights by examining concentrations of glucose, insulin, and other factors involved in glucose metabolism.
Obstructive Sleep Apnea and Metabolic Health
Sleep; ApneaMetabolism DisorderThis is an observational study involving chart review of patients seen in the bariatric clinic since 2012 at The Ohio State University (OSU). Obese patients will be divided into two groups according to their metabolic profile (healthy vs unhealthy). The assignment to each group will be determined by the presence of diagnosis of hypertension, diabetes mellitus and/or dyslipidemia. The presence of at least 2 diagnoses will be defined as metabolically unhealthy. Then, each group will be divided based on the presence of Obstructive Sleep Apnea diagnosis. All data will be collected through a review of the patient's electronic medical record from the bariatric clinic database (the investigators will collect variables as age, gender, race, BMI, smoking status, the presence of diagnosis as Obstructive sleep apnea, hypertension, diabetes, hyperlipidemia). The investigators will analyze that data and establish if there is any correlation between sleep apnea and the subjects' metabolic profile.

Effects of Exercise and Energy Balance on 24-hour Blood Glucose Control
Metabolic DiseaseAdipose TissueThe study team will assess "free-living" 24h glucose control using continuous glucose monitors (CGM) over an 88h period on two separate occasions. Importantly, standardized meals will be provided and will be consumed at specific times each day of the CGM measurement period. These meals will be identical on day 1 and day 3 of measurement. Each subject will complete two CGM periods, which will be identical except for the calorie and macronutrient content of the post-exercise dinner. In addition, during trial 1, two CGM's will be worn, while only 1 CGM will be worn on trial 2.
Endogenous Glucose Production in Patients With Glycogen Storage Disease Type Ia
Glycogen Storage Disease Type IAGlycogen storage disease type Ia (GSDIa) subjects retain a limited capacity for endogenous glucose production (EGP). To date, the origin of residual EGP in GSDIa patients is unknown. Either increased glycogen debranching or lysosomal glycogen breakdown can account for residual EGP in GSDIa. Innovative treatments for GSDIa (e.g. AAV8-mediated gene therapy and mRNA therapy) are being developed.Therefore, longitudinal minimally-invasive monitoring of outcomes after therapeutic interventions in GSD Ia subjects becomes warranted. The primary objective is to test the feasibility of EGP quantification in adult GSDIa subjects by stable isotopes after a single oral [6,6-2H2]glucose dose. Secondary objectives are to compare EGP assessed by a single oral [6,6-2H2]glucose dose (a) in GSDIa patients versus matched healthy participants, (b) among GSDIa patients, (c) in the pre-prandial state versus the fed state, (d) in the controlled hospital setting versus the home setting. Data collected from the continuous glucose monitoring data will also be compared
