Paclitaxel Detection in NSCLC Treated With TC Regimen
Non-small Cell Lung CancerBy detecting the blood concentration of paclitaxel (PTX), Investigator assume this research can identify the individual differences of PTX pharmacokinetics (PK) parameters (TC>0.05 refers to the duration of paclitaxel plasma concentration above 0.05 µmol/L) in Chinese non-small cell lung cancer (NSCLC) patients, and find the correlation between PK results and PTX toxicities and Effectiveness, acquire the optimization method of PTX, and finally try to explore the individualized PTX pharmacokinetically-guided dosing strategy. Orally administer rosiglitazone, which is a substrate of CYP2C8 the same as paclitaxel, before chemotherapy injection. Detect the blood concentration of rosiglitazone, analyze the correlation of rosiglitazone pharmacokinetic parameter and paclitaxel exposure, and explore the effect of rosiglitazone as an in vivo probe of paclitaxel exposure. The variability of paclitaxel concentrations in the patient population dosed by body surface area (BSA), and the limitation of BSA-based dosing of paclitaxel. Verify that paclitaxel TC>0.05 is the most relevant predictor of haematological toxicity and clinical outcomes. Lung NeoplasmsBackground: Anetumab ravtansine is a new drug. It kills cancer cells that carry mesothelin. That is a protein on the surface of tumor cells in many types of tumors, including most lung cancers. Researchers want to find a safe dose for the study drug for lung cancer. They want to see if it can shrink tumors in mesothelin-positive lung cancer. Objectives: To test the safety and effectiveness of anetumab ravtansine for lung cancer. Eligibility: Adults 18 years and older who have lung cancer that has gotten worse on other therapy Design: Participants will be screened with: Medical history Physical exam Tumor tissue sample. This can be from a previous procedure. Blood and urine tests Heart tests Scans. For one scan, a small amount of radioactive substance is injected into the blood. Eye exam The study will have 21-day cycles. On day 1 of each cycle, participants will get the study drug through a tube inserted in a vein. Participants will repeat a heart test in cycles 1 and 2. They will have blood tests weekly in cycle 1, twice in all other cycles. They will have scans every 6 weeks for the first 6 months, every 9 weeks until the end of year 2, then every 12 weeks. Participants will have samples of tumor tissue taken twice. About 30 days after stopping the study drug, participants will have a follow-up visit. This will include medical history, physical exam, blood and pregnancy tests, and heart and eye tests. Some will be called a few times a year to discuss their health and treatment.

Squamous Cell Lung CancerDasatinib is a drug that has been shown to stop some cancer cells from growing. This drug has been used in treatment for other types of cancer and information from other research studies suggests that dasatinib may help to stop squamous cell lung cancer from growing, especially in individuals whose tumor has a mutation in the DDR2 gene. Advanced squamous cell lung cancer (SqCC) carries a poor prognosis and new therapeutic targets are needed. Several studies have examined dasatinib in NSCLC; these report significant toxicities, but also responses in patients found to harbor mutations in DDR2 or BRAF. An open-label phase II trial with dasatinib was carried out to determine the response rates in patients with SqCC who had previously failed standard chemotherapy and to correlate responses with patient genotype.

Imatinib mesylate may stop the growth of tumor cells by blocking some of the enzymes needed for cell growth. Giving gemcitabine together with imatinib mesylate may kill more tumor cells. PURPOSE: This phase II trial is studying how well giving gemcitabine together with imatinib mesylate works in treating patients with recurrent or metastatic non-small cell lung cancer.
